The effect of different treatments of leaf and stem explants and hormones with different concentration on callus weight, the number of shoots and regeneration of Thymus daenensis

Thymus daenensis is a plant from Lamiaceae family that is a species endemic to Iran for several years. Infusion of the aerial parts of thymus species is tonic, carminative, antispasmodic, anti-inflammatory, expectorant which is used as traditional cold remedy. According to the exposure of the plant due to the indiscriminate use as a medicinal plant and and protection from natural resources, T. daenensis tissue culture were done to increase the quality and quantity of its constituents through callus as well as micropropagation to multiply and produce a large number of progeny Thymus by using hormonal different treatments on leaf and stem explants. MS medium with different concentrations of naphthalene acetic acid, benzyl amino purine, kinetin and 2,4-D (mg/l) of callus formation was observed. Based on the results of interaction of various explants and hormone treatments with different concentrations, the highest callus weight (87/1 g) was related to the treatment of 0.2 NAA+1.5 BAP mg/l and leaf explants. Furthermore, the results illustrated that the highest number of shoots (33) related to the treatment of 0.8NAA + 6 BAP mg/l and was for the stems. Also the highest number of regeneration (10) was reported in leaf and the stems was treated with 0.2 NAA + 1.5 BAP mg/l. Results of the callusing and regeneration of T. daenensis to enhance the quality and quantity of callus and produce a large number of progeny plants showed that the effects different explants and hormones with different concentrations and as well as their interactions on the number of shoots, regeneration and callus weight was significant at level of 1 %.
