Morphine down regulates Kisspeptin/GPR54 signaling system in male rats

Opioids exert inhibitory effects on reproduction dominantly via inhibiting the GnRH/LH release. kisspeptin signaling pathway play a role in relaying the environmental and metabolic information to reproductive axis. In the present study the effects of morphine was investigated on kisspeptin/GPR54 signaling system activity. Twenty Wistar male rats weighing 230-250g in four groups (in each group n=5) received saline, morphine(5mg/kg), naloxone (2mg/kg) or naloxone/morphine respectively. Hypothalamic samples were dissected and mean relative KiSS1or GPR54 mRNA levels were determined RT-PCR. Morphine or naloxone caused a significant decrease or increase in mean relative KiSS1 gene expressions compared to saline respectively. Injections of naloxone plus morphine increased significantly the mean relative Kiss1 gene expression compared to morphine. While mean relative GPR54 gene expressions did

not changed following morphine or naloxone injections compared to saline. Down regulation of Kiss1 gene expression may be partly involved in the mediating the inhibitory effects of morphine on reproductive axis.
