Investigation of the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of Sambucus ebulus methanolic extract on Gram Positive and Gram Negative Bacteria “in Vitro”

Bacterial infections are one of the most important problems of health system in all countries. Due to complications and side effects of chemical drugs and increasing bacterial resistance to these drugs, this study aimed at the effects of Sambucus ebulus on Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa. Mentioned strains are important factors of nosocomial infections and are resistant to many antibiotics. Methanolic extract of studied plant prepared by maceration method and phenol and Flavonoid contents were measured using Folin Ciocalteau and aluminum chloride methods, respectively. Inhibitory zone was studied using well diffusion method. Antibiotic disks such as tetracycline, Gentamicin and Ciprofloxacin were laid to control. Minimum inhibitory concentration of micro organisms (MIC) and minimum bactericidal concentration of micro organisms (MBC) was determined using broth dilution method. The entire process for the extract was performed in triplicate. All data were analyzed using the software SPSS_21. Inhibitory zone diameter for Sambucus ebulus on Gram negative Klebsiella pneumoniae bacteria had the most anti microbial effect with 5.7 mm, and recorded the lowest inhibitory zone diameter for Gram negative Pseudomonas aeruginosa bacteria with 2.3mm. Sambucus ebulus methanolic extract was not effective as much as it could be offered as a new source of anti microbial herbal medicine. This study was performed at extreme concentration of 400μg/ml and the anti microbial effect dose dependently increased, so this effect may continuously increased if more concentrated plant extract is used.
